There are twow ways to save your game. One is to reach certain points in the Main Case and it asks if you wish to saves. The other is using the restrooms around the mall.

I feel your pain man. I was doing the saint play through and forgot to complete 1 case where Isabella tells you the cure and I couldnt save the last survivor I needed. And dealing with those damn AI is maddening. All ruined by a poorly timed save. Just have to take a deep breath and start again.
